摘要
目的建立测定血浆中头孢拉定浓度的方法。方法采用HPLC法。用Eclipse XDB—C8色谱柱(150mm×4.6mm,5μm),以乙腈-磷酸盐缓冲液(17:83)为流动相,N-对甲氧基苯甲酰基丁酸(ABA)为内标,于254nm测定。结果头孢拉定在0.5~30.0μg·ml^-1浓度范围内线性良好,最低定量浓度为0.5μg·ml^-1,日内RSD〈8%,日间RSD〈13%,平均相对回收率99.1%。结论所建方法操作简单、快速、重复性好、准确可靠,可用于临床血药浓度的监测及药动学的研究。
OBJECTIVE To establish a method for the determination of the concentration of Cefradine in plasma.METHODS HPLC was used to determine Cefradine in human plasma,with the Eclipse XDB - C18 column (150 mm× 4.6mm,5μm)as analytical column and the mobile phase consisting of acetonitrile - phosphate buffer solution( 17 : 83, pH4). The internal standard was 4 - p - anisamidobutyric acid (ABA) (20μg·ml^-1 diluted by methanol), with the detective wavelength of 254 nm.RESULTS The linear range of Cefradine was 0.5 - 30.0μg·ml^-1. The average recovery was 99.1% with RSD 〈 8 %. CONCLUSION The method is simple, rapid, accurate, precise and repeatable. It is applicable to determine the concentration of Cefradine in human plasma for pharmacokinetic studies.
