Determination of Cefradine in plasma by HPLC

目的建立测定血浆中头孢拉定浓度的方法。方法采用HPLC法。用Eclipse XDB—C8色谱柱（150mm×4．6mm，5μm），以乙腈-磷酸盐缓冲液（17：83）为流动相，N-对甲氧基苯甲酰基丁酸（ABA）为内标，于254nm测定。结果头孢拉定在0.5～30．0μg·ml^-1浓度范围内线性良好，最低定量浓度为0．5μg·ml^-1，日内RSD〈8％，日间RSD〈13％，平均相对回收率99．1％。结论所建方法操作简单、快速、重复性好、准确可靠，可用于临床血药浓度的监测及药动学的研究。

OBJECTIVE To establish a method for the determination of the concentration of Cefradine in plasma.METHODS HPLC was used to determine Cefradine in human plasma,with the Eclipse XDB - C18 column （150 mm× 4.6mm,5μm）as analytical column and the mobile phase consisting of acetonitrile - phosphate buffer solution（ 17 ： 83, pH4）. The internal standard was 4 - p - anisamidobutyric acid （ABA） （20μg·ml^-1 diluted by methanol）, with the detective wavelength of 254 nm.RESULTS The linear range of Cefradine was 0.5 - 30.0μg·ml^-1. The average recovery was 99.1% with RSD 〈 8 %. CONCLUSION The method is simple, rapid, accurate, precise and repeatable. It is applicable to determine the concentration of Cefradine in human plasma for pharmacokinetic studies.
