摘要
目的探讨利用HER-2阳性细胞系SKBr-3建立自发性乳腺癌肝转移模型,为乳腺癌肝转移的机制及治疗学研究提供有价值的模型。方法利用SKBr-3乳腺癌细胞株接种至16只裸鼠第二对乳腺脂肪垫,建立裸鼠第一代原位乳腺癌模型,再利用建立成功的原位肿瘤组织在16只裸鼠原位进行瘤组织块第二代原位种植,最后利用第二代组织块种植成功的瘤组织对16只裸鼠再次进行原位肿瘤种植,分别观察肿瘤肝转移情况。结果在第一代肿瘤细胞原位种植的肿瘤中,没有发生肝转移现象。而在第二代组织原位种植瘤中,只有1只发生肝转移,而在第三代的组织原位肿瘤种植模型中出现了接近81.25%(13/16)的肝转移。无一只出现肉眼肺转移。结论乳腺癌裸鼠体内反复传代可以建立高效、相对特异性的自发性乳腺癌肝转移模型,为肝转移机制及治疗研究提供很好的模型。
Objective To establish a mouse model of orthotopic breast cancer with liver metastasis using HER - 2 posi- tive cell line SKBr- 3 and to provide a valuable model for study in mechanism and treatment of breast cancer with liver metastasis. Methods The first generation orthotopic nude mice breast cancer model was established by injection of SKBr- 3 cell line into second mammary fat pads of 16 nude mice. Intact tumor grafts from the first generation mouse model were inoculated orthotopitally into another 16 nude mice to establish the second orthotopic nude morse breast cancer model. The procedures were repeated to establish the third generation model. Finally, lung and liver metastasis was evaluated under micmscepic and macroscopic observation. Results There was no macrometastasis in the first generation mouse model either in the lung or in the liver and liver metastases was only found in one animal in second generation mouse model. Howerver,liver macrometastases was found in 13/16 (81%) animals without lung macrometastasis. Conclusion A highly efficient and more specific orthotopic breast cancer liver metastatic mouse model can be established by repeated grafting of tumor tissues into nude mice and can be an excellent model in studying the mechanism and treatment of breast cancer with live metastasis.
