摘要
目的 观察家兔实验性动脉粥样硬化(atherosclerosis,AS)形成过程中心肌Ⅰ,Ⅲ型胶原的动态变化。方法 雄性新西兰家兔40只,随机等分为正常对照组(C组)和AS模型组(AS组)。分别于实验的第4,8,12周末随机处死每组家兔各5~8只,定位取带有冠状动脉主干及分支的心脏组织;常规石蜡切片,行HE染色及天狼猩红染色;偏光镜下观察胶原成分的分布及其量的变化。结果 与C组相比,4周末时.AS组兔心肌细胞间隙和心肌间小血管周围两型胶原已见增多、增粗;8周末以后,增加更加明显,尤其是Ⅰ型胶原。结论 AS形成过程中,心肌胶原呈持续增高趋势,且以Ⅰ型胶原为主。
Objective To study the dynamic changes of type Ⅰ and Ⅲ collagen of myocardium interstitium in rabbits developed atherosclerosis. Methods 40 male New Zealand rabbits were equally randomly divided into 2 groups: control group (C group) and atherosclerosis model group (AS group). At the 4th, 8th, 12th weekends of the experiment, 5 - 8 animals in each group were sacrificed, and cardiac tissue with coronary arteries were harvested for paraffin sections. Sections were stained with hematoxylin - eosin and picro - sirinsred separately. The content and distribution of type Ⅰ and Ⅲ collagen were examined with polarization microscopy. Results Comparing with the C group, the content of beth types of collagen in AS group significantly increased in the interstitial tissue at 4^th weekend; after 8^th weekend, this phenomenon became more serious, especially for type Ⅰ collagen. Conclusion The content of both types of collagen tends to increase persistently as athrosclerosis develops. The increase of type Ⅰ collagen is more distinct than that of type Ⅲ collagen.
