摘要
目的观察活血复方对大鼠缺血再灌注模型神经行为学和大脑皮质及海马神经细胞凋亡的影响。方法采用右侧颈总动脉夹闭缺血再灌注手术,建立大鼠脑缺血再灌注及神经细胞凋亡模型,并观察大鼠神经行为学改变,HE染色光镜观察大脑皮质及海马病理形态改变,在缺血再灌注不同灌注时相TUNEL法检测神经凋亡细胞,并定量分析凋亡相关基因bcl-2及bax的变化,以及活血复方对脑缺血再灌注神经细胞凋亡的影响。结果缺血再灌注组神经行为学计分较假手术组为高;皮质及海马神经细胞出现退化、变性、坏死等病理变化;术后6h皮质及海马细胞神经细胞凋亡百分比升高。活血复方能有效改善大鼠神经行为症状,降低皮质及海马神经细胞凋亡百分比,增加bcl-2表达,减少bax表达,与模型组相比具有显著差异。结论活血复方可以减缓缺血再灌注后皮质及海马神经细胞凋亡的影响,较好地改善大鼠脑缺血再灌注模型的神经行为症状。
Objective:To investigate the effect of Huoxue Recipe(HR,活 血 复 方 ) on neurogenic behavior and hippocampal nerve cell apoptosis in rats with cerebral ischemia/reperfusion.Methods:Rats model of cerebral ischemia/reperfusion injury was established by right common carotid artery blocking.The neurogenic behavior of rats was observed,pathological changes of cortex and hippocampus were observed with HE staining and light microscope,morphology of apotosis nerve cell was observed by fluorescence microscope and percentage of apoptotic cell was measured by TUNEL method.Meanwhile,the quantitive assessment of apoptotic relative gene bcl- 2 and bax was made.Result:The score of neurogenic behavior of cerebral ischemic- reperfusio rats was higher than that of the sham operated group.Pathological changes such as cortical and hippocampal nerve cell regression,degeneration,and necrosis were found in the model rats,percentage increase of hippocampal nerve cell reached the peak after 6 hours of the operation.The experiment showed,HR could effectively improve the neurogenic function,lower the percentage of apoptosis significantly compared with those in the cerebral ischemic- reperfusion group,bcl- 2 positive protein cells were increasing while bax positive protein cells declining significantly compared with the the cerebral ischemic- reperfusion group.Conclusion:HR has the effect of improving the neurogenic behavior,reducing the cell apoptosis rate of cortex and hippocampus in mice with ischemia- reperfusion injury.
