基质金属蛋白酶-7在子宫内膜癌中的表达

Expression of Matrix Metalloproteinase-7 in Human Endometrial Carcinoma

目的:观察基质金属蛋白酶-7(m atrixm etalloprote inase-7 MMP-7)在正常子宫内膜、子宫内膜非典型增生及子宫内膜癌组织中的表达及其与子宫内膜癌组织学分级和临床分期的关系。方法:检测38例子宫内膜癌组织,7例子宫内膜非典型增生组织及10例正常子宫内膜组织中MMP-7的蛋白定位及mRNA表达水平并结合临床病理特征进行分析。结果:MMP-7蛋白主要定位于子宫内膜腺上皮细胞及肿瘤细胞中;子宫内膜非典型增生组织中MMP-7表达水平与正常组织无显著差异;子宫内膜癌中MMP-7表达明显增高(P<0.05),并与癌组织手术-病理分期和组织学分级密切相关。结论:MMP-7参与子宫内膜癌的侵袭和转移,有望成为子宫内膜癌发展及预后判断的一种新的参考指标。

Objective： To observe the expression of matrix metalloproteinase-7 （MMP-7） in normal, premalignant and malignant endometrial samples and its relationship with histopathological grades and clinical stages in endometrial cancer patients. Methods： A total of 38 cases of endometrial carcinoma samples, 7 of premalignant samples and 10 of normal endometrial tissue were tested for expression of MMP-7 by immunohistochemistry and RT-PCR. Results： Immunostaining localized MMP-7 primarily in epithelial glandular cells and tumor ceils. Semi-quantifications with immunohistochemistry and RT-PCR revealed that the expression of MMP-7 was not different between normal endometrial samples and premalignant group. In contrast significantly higher leveh were found in the malignant tissues, and MMP-7 expression was positively related to histopathological grades and clinical stages. Conclusions： Increased expression of MMP-7 in advanced tumors suggests its role in tumor development and tumor metastasis. These results provide a new significant marker in the judgment of tumor invasion or metastasis and the estimate of prognosis with cndometrial carcinoma.
