脑出血后抑郁模型大鼠脑内5-羟色胺、去甲肾上腺素和酪氨酸羟化酶的表达 被引量:4

Expressions of hydroxytryptamine,norepinephrine and tyrosine hydroxylase in brain of model rats with post cerebral hemorrhage depression

摘要 目的:观察脑出血后抑郁动物模型脑内单胺神经递质5-羟色胺、去甲肾上腺素和酪氨酸羟化酶的表达变化,分析脑出血后抑郁症的发生机制。方法:实验于2004-02/2005-01在北京中医药大学科研中心进行,健康Wistar雄性大鼠32只,随机分为正常组,脑出血组、抑郁组、模型组,每组8只,正常组常规喂养;脑出血组仅进行脑出血手术,术后常规喂养;抑郁组正常饲养7d后,予21d的慢性应激;模型组先进行脑出血手术,术后常规喂养7d后,予21d的慢性应激。应激结束后,采用免疫组织化学方法观察脑出血后抑郁模型大鼠脑内5-羟色胺、去甲肾上腺素、酪氨酸羟化酶的表达改变。结果:各组大鼠全部进入结果分析。①脑内5-羟色胺阳性细胞的表达:模型组大鼠脑海马CA1、CA3、CA4、梨状皮质、扣带皮质、下丘脑内5-羟色胺阳性细胞表达减少,颜色浅淡,与正常组比较有统计学显著性意义。脑出血组5-羟色胺阳性细胞在海马CA3、CA4、梨状皮质、扣带皮质、下丘脑数量表达减少,与正常组比较有统计学显著性差异。慢性应激抑郁组在海马CA1、CA3、梨状皮质、扣带皮质、下丘脑5-羟色胺阳性细胞表达减少,与正常组比较有统计学显著性差异。②酪氨酸羟化酶和去甲肾上腺素的表达:模型组、出血组、抑郁组下丘脑内酪氨酸羟化酶阳性细胞表达减少,颜色变浅,与正常组比较有统计学差异。脑出血组、抑郁组、模型组去甲肾上腺素阳性细胞在扣带皮质和下丘脑内颜色变浅,呈空泡样,较正常组明显减弱(P<0.05)。结论:脑出血后抑郁症模型大鼠脑内5-羟色胺和去甲肾上腺素和酪氨酸羟化酶表达下降。 AIM: To observe the changes of expressions of the monoamlne neural ,transmitters including hydroxytryptamine (5-HT), norepinephrine (NE) and tyrosine hydroxylase (TH) in the brain of model rats with post cerebral hemorrhage depression, in order to find the pathogenic mechanism of post cerebral hemorrhage depression. METHODS: From February 2004 to January 2005 ,the experiment was conducted in the Research Center of Beijing University of Traditional Cbinese Medicine. Totally 32 male Wistar rats were randomly divided into 4 groups: normal group, hemorrhage group, depression group and model group with 8 rats in each group. The rats in the normal group were fed regularly; the rats in the hemorrhage group were also fed regularly after cerebral hemorrhage operation; the rats in the depression group were given chronic stress for 21 days after 7 days of normal feeding; the rats in the model group were performed cerebral hemorrhage operation and given chronic stress for 21 days after 7 days of normal feeding. After the stress, the changes of the expressions of 5-HT, NE and TH in the brain were observed with immunohistochemical method. RESULTS: All the 32 rats were involved in the result analysis. (1)The expressions of 5-HT positive cells in brain: The expressions in the model group decreased in the areas of CA1, CA3 and CA4 in hippocampus, piriform cortex, cingulate cortex and hypothalamus, which had significant differences from the normal group, and the cells turned lighter color. The expressions in the hemorrhage group decreased in CA3 and CA4 of hippocampus, piriform cortex, cingulate cortex and hypothalamus, which had significant difference from the normal group. The expression in the depression group decreased in CAI and CA3 of hippocampus, piriform cortex, cingulate cortex and hypothalamus, which also had significant differences from the normal group, (2)The expressions of TH and NE: The TH positive cells in the hypothalamus of the model group, hemorrhage group and depression group decreased and turned lighter color, which had significant differences from the normal group. The NE positive cells in the cingulate cortex and hypothalamus of the model group, hemorrhage group and depression group showed vacuolar-shape and turned lighter color, which were obviously weaker compared with the normal group (P 〈 0.05). CONCLUSION: The decreased rule of the expressions of 5-HT, NE, TH in the brain of model rats with post hemorrhage depression is in accordance with that of the center monoamine neural transmitters in patients with post-stroke depression in clinic.
