丝羽乌骨鸡mtDNA限制酶酶切研究

Restriction Study on mtDNA from Silkies

应用碱变性法制备丝羽乌骨鸡（简称丝羽鸡）肝细胞ｍｔＤＮＡ，测其分子量为１８．５ｋｂ．用限制性内切酶进行酶切分析，结果表明，ＢａｍＨⅠ、ＰｓｔⅠ、ＨｉｎｄⅢ、ＳａｃⅠ在丝羽鸡ｍｔＤＮＡ分子上分别有１、１、１、２个酶切位点．根据单酶和双酶完全酶切片段的分子量，建立了丝羽鸡ｍｔＤＮＡ的限制酶图谱．与由来亨鸡肝细胞ｍｔＤＮＡ为材料所得的研究结果比较，发现丝羽鸡与来亨鸡在ｍｔＤＮＡ种质方面存在着较大差异．

mtDNA from liver of Silkies was prepared by the alkaline lysis procedure. its molecular weight was measured to be 18. 5 kb, The results show that restriction endonucleaesBamH I, Pst I,Hind Ⅲ and Sac I have 1, 1, 1, 2 sites on the mtDNA molecule, respectively, uslug analysis of restriction enzyme cleavage. The restriction map of mtDNA from Silkies was constructed,according to the data of moleculat weight of generated fragments by complete digestion with single and double enzyme. mtDNAs of liver from Silkies and Leghorns have heredity variatin as compared with study results from Leghorns.
