The structure types, pharmacological activities and clinical development of peripherally restricted kappa opioid receptor agonists recently reported were reviewed. Peripheral kappa opioid receptor has been identified in different tissues outside the CNS in animal and human. Its agonists are a class of potent analgesic and antiinflammatory agents, which selectively bind to peripheral kappa opioid receptors, thereby avoiding morphine-like and central side effects.
