摘要
氨基酸含量分析是蛋白质化学研究的重要手段之一,样品前处理方法已成为当前影响测定氨基酸含量精确度的首要问题。本文研究添加不同保护剂对水解鸡毛样品的氨基酸影响。A法:0.2%硫代乙醇酸的6mol/LHCl;B法:0.2%硫代双乙醇的6mol/LHCl;C法:6mol/LHCl、实验证明:B法水解测定的氨基酸总量高于其他两种方法;B法测定的必需氨基酸也高于其他两种方法。所以,B法是水解羽毛角蛋白较理想的前处理方法。
Quantitative analysis of amino acid is one of the important studies on protein chemistry. Hydrolysis of samples is a main effect on quantitative analysis of amino acid. A reseach is made for effect of adding different pretective agents to 17 kinds of amino acids in hydrolyzed chicken feather samples.(A: 0. 2%thioglycolic acid 6mol/L HCl;B: 0. 2% thiodiglycolic acid 6mol/L HCl;C: 6mol/L HCl) The result showed that B pretective agent would get more total amiono acid and necessary amino acid than A or C agent.
