摘要
研究用于癌症诊断与治疗的光敏剂血卟啉(hematoporphyrin derivative,HPD)对人体癌细胞与正常细胞的亲和性,采用超短脉冲激光光谱技术和皮秒时间相关单光子计数系统,测量经血卟啉培养的活体癌细胞与正常细胞的荧光光谱,观测到癌细胞样品在645nm处具有特有的光谱谱峰;测量不同浓度血卟啉荧光光谱,观测到随着血卟啉浓度的逐渐增大,在645nm处出现的光谱峰也随之显著增强。对荧光光谱的二阶导数光谱进行分析,发现癌细胞与浓度2×10-1mg/ml血卟啉的二阶导数光谱图形状相似,说明在癌细胞内部血卟啉浓度增大了约2个数量级。测量数据表明癌细胞对血卟啉有很强的亲和性,同时还得到了亲和性的量化值。测量结果确认了荧光光谱技术诊断与治疗癌症的可行性,并对化疗注射光敏剂药液浓度的计量提供参考。
The object of this study is to cancerous and normal cells intimacy of hematoporphyrin derivative (HPD) for diagnosis and therapy cancer, fluorescence spectra of cancerous and normal cells are measured using a ultrashort pulses laser spectral technique and picosecond time- correlated single- photon counting system. Characteristic fluorescence spectra peak near 645nm of cancerous cells sample are observed, influence of HPD concentration is also presented on the characteristic fluorescence spectra peak near 645nm, second derivative spectra are obtained by calctdating date of fluorescence spectra, second derivative spectra figure of cancerous cells is approximately with HPD 2 × 10^-1 m g/ml, HPD concentration is increased two order of magnitude in cancerous cells, the data show cancerous cells is greatly intimate with HPD. The results obtained can be used as an important basis for diagnosis and therapy cancer based on ultrashort pulses laser spectral technique, the results will contribute to metrology of HPD concentration in therapy cancer.
