摘要
目的:通过观察硝苯地平(NIF)对牙龈上皮细胞增殖和凋亡的影响,探讨硝苯地平致龈增生的机理。方法:采用MTT法检测不同浓度NIF对体外培养的人牙龈上皮细胞增殖的影响;采用流式细胞仪(FCM)检测上皮细胞受NIF诱导后凋亡小体形成百分率。结果:24 h、48 h7、2 h后各加药组与对照组之间MTT值没有明显差异;48 h后高浓度药物组(1200μg/l)凋亡小体形成率小于对照组(P<0.05),且主要作用于细胞G0/G1期和S期。结论:硝苯地平对体外培养的人牙龈上皮细胞的增殖没有直接刺激作用,但高浓度的NIF可抑制细胞凋亡。
Objective:To observe the effect of NIF on proliferation and apoptosis of gingival epithelial cells. Method: Culture human giugival epithelial cells in vitro. Observe the effect of different concentration of NIF on proliferation of gingival epithelial cells through evaluating MTT value, and analyze the effect of different concentration of NIF on apoptosis of gingival epithelial cells by flow cytometry. Result: the MTT values have no difference among the test groups and the control after 24,48,72 hours(P〉0.05). After 48 hours the concentration of 1200 μg/1 group has a lower apoptosis than the control(P〈0.05), And the effect occurs in G0/G1 phase and S phase. Conelusion:NIF cannot directly stimulate the proliferation of cultured human gingival epithelial cells. But high concentration of NIF can inhabit apoptosis of gingival epithelial cells.
