大鼠舌癌变过程中NF-κB和cyclinD1蛋白的表达 被引量:1

Changes in nuclear factor κB and cyclinD1 expression during 4-nitroquinoline 1-oxide induced rat tongue carcinogenesis

摘要 目的:探讨大鼠口腔癌变过程中核转录因子κBp65(NF-d3p65)、cyclinDl蛋白的表达及其生物学意义。方法:0.002%4-硝基喹啉-1-氧化物(4NQO)饮水喂养Wistar大鼠9~32周建立大鼠舌癌变模型,用免疫组化sP法检测38例大鼠舌癌变过程不同病理阶段组织中NF-d3P65、cyclinD1蛋白的表达。结果:常黏膜、轻度异常增生、中度异常增生、重度异常增生、原位癌、鳞癌组织中,NF-d3的阳性率分别是20%、20%、50%、62.5%、50%和83.33%,(鳞癌与正常黏膜组织中NF-d3P65的表达有显著性差异(P〈0.05);cyclin D1阳性率分别是20%、60%、62.5%、87.5%、100%和83.33%,重度异常增生、原位癌、鳞癌组织中cyclin D1表达显著高于正常黏膜(P〈0.05或P〈0.01)。两者的表达均与组织的病理学分级相关(P〈0.01),两者的表达在鳞癌阶段具有相关性(P〈0.05)。结论iNF-κBp65、cydinDl蛋白表达与口腔癌发生发展相关,NF-d3通路在口腔黏膜癌变过程起重要作用,NF-κB、cyclinD1可能成为口腔癌变病变生物标志物之一。 Objective:To observe the different expression of NF-κBP65 and cyclinD1 during oral carcinogenesis, analyze the relationship between the abnormal expression of NF-κBp65, cyclinD1, and the occurrence and development of oral carcinogenesis. Method.S-P immunohistochemlcal method was employed to detect the expression of NF-κBp65 protein and cyclinD1 protein in 38 rat tongue carcinogenesis specimens induced by 4-nitroquinoline 1-oxide. Result: with the progress of tongue carcinogenesis, the expression of NF-κBp65, cyclinD1 was up-regulated, in normal, Mild epithelial dysplasia, Moderate epithelial dysplasia ,severe epithelial dysplasia ,carcinoma in situ and squamous cell carcinoma(SCC), The positive rate of NF-κBp65 was 20% ,20% ,50% ,62.5% ,50% 和83.33%, respectively. ( there was significant di{ferences between normal and SCC, P 〈 0.05) ; while that of cyclinD1 was20 %, 60 %, 62.5 %, 87.5 %, 100 % and 83.33 %, respectively. ( there was significant differences between normal and severe epithelial dysplasia ,carcinoma in situ and SCC, P〈 0.01 or P 〈 0.05). There was a significant correlation between the increased levels of NF-κBp65, cyclinD1 and histopathological grade. The positive expression of NF-κBp65 was also associated with that of cyclinD1 in SCC. Conclusion: The up expression of NF-κBp65 and eyclinDl protein may be correlated to the occurrence and the development of oral carcinoma, activated NF-κB plays an important role in the overexpression of cycllnD1. Furthermore, NF-κB and cyclinD1 may be the useful biomarker of oral precancerous lesion.
