摘要
目的探讨应用重组人粒细胞集落刺激因子(rHuG-CSF)动员外周血干细胞时,异体、自体造血干细胞移植供、受者的骨髓及外周血CD3+4细胞和T细胞亚群的动态变化特点及最佳外周血干细胞采集时间。方法对兰州军区兰州总医院2001~2002年间的12名健康供者和16例化疗7d后的恶性血液病患者,以rHuG-CSF每12h150μg,皮下注射,连用5d。应用流式细胞仪测定骨髓及外周血CD3+4细胞和T细胞亚群。结果(1)应用rHuG-CSF前,两组的骨髓CD3+4细胞差异有显著性意义(P<0·01)。应用48h以后,两者的骨髓CD3+4细胞均较前有明显增加(P<0·01),两组之间差异仍存在显著性意义(P<0·01)。两者的骨髓CD3+4细胞高峰值出现在96h后,较应用前差异有显著性意义(P<0·01)。(2)应用rHuG-CSF72h以后,两组外周血中CD3+4细胞均较前有明显增加(P<0·01)。96h以后,两组的CD3+4细胞达高峰,较应用前差异有显著性意义(P<0·01)。(3)应用rHuG-CSF对两者的T淋巴细胞亚群无明显影响。结论应用rHuG-CSF动员外周血干细胞96h以后,异体、自体造血干细胞移植供、受者的骨髓及外周血CD3+4细胞均达到高峰,可适时采集外周血干细胞。
Objective To approach the dynamic changes of CD34^+ cells and T lymphocyte subsets and best time of peripheral blood stem cell harvest when using rHuG-CSF for peripheral blood stem cell mobilization in donors and patients. Methods From 2001 to 2002 12 donors and 16 patients in Lanzhou General Hospital who received chemotherapy 7 days ago were injected rHuG-CSF 300μg/d for mobilization peripheral blood stem cells, and flow cytometry were used to detect the changes of CD34^+ cells and T lymphocyte subsets for 5 days. Results (1)Betore using rHuG-CSF, there was obvious difference between patients and donors in bone marrow CD34^+ cells (P 〈 0.01 ) . After using for 48h there was obvious difference ( P 〈0.01 ) . After 96 h the highest values were reached. There was significant difference ( P 〈0.01 ) . (2) After using rHuG-CSF for 72 h, there was obviously increase in peripheral blood CD34^+ cells in patients and donors ( P 〈0.01 ) . After 96 h the highest values were reached. There was significant difference ( P 〈0.01 ), (3) There were no obvious changes in the donors' and patients' T lymphocyte subsets when using tHuG-CSF for some days. Conclusion After mobilization for 96 h, the higest values of CD34^+ cells in bone marrow or peripheral blood of patients and donors ware reached and peripheral blood stem cells may be harvested.
