摘要
目的:探讨二烯丙基三硫(diallytrisulfide,DATS)诱导人胃癌细胞系MGC-803凋亡与细胞内活性氧(re-activeoxygenspecies,ROS)之间的关系。方法:用12mg/L的DATS作用于体外培养的MGC-803细胞24h后,用流式细胞术检测凋亡率及细胞内活性氧水平。结果:12mg/LDATS作用于MGC-803细胞24h后,细胞凋亡率明显上升;细胞内活性氧水平与DATS呈浓度依赖性升高。用抗氧化剂NAC预处理细胞,能抑制细胞内活性氧水平的升高及DATS诱导的细胞凋亡。结论:DATS诱导胃癌细胞凋亡与细胞内活性氧产生增加有关。
Objective: To explore the relationship between the effect of DATS inducing apoptosis of MGC-803 cells and the intraeellular level of reactive oxygen. Methods: Flow eytometry was used to detect the intraeellular reactive oxygen species (ROS) level and the rate of apoptosis. Results: MGC- 803 cells treated with DATS significantly increased in the intracellular ROS level and the percentage of apoptosis cell. Pretreatment of N-aeetyl-1-eystein, a thiol-eontaining antioxidant effectively attenuates DATS-induced increasing of intracellular ROS and the consequent apoptotic of MGC803. Conclusion: These results suggest that the cellular generation of ROS plays a role in initiating DATS-mediated apoptosis of MGC-803 cells.
