The Effect of Radionuclide Antisense Therapy Targeting Ki67 Gene on Proliferation and Apoptosis of the Human Renal Carcinoma Cells in vitro and in vivo

摘要 目的:探讨Ki-67基因放射性核素反义治疗对人肾癌细胞生长及凋亡的影响。方法:125I标记的反义核酸(125I-ASODNs)处理人肾癌786-0细胞及荷瘤小鼠,采用免疫组化、Westernblot检测Ki-67表达;TUNEL法检测细胞凋亡。结果:肾癌细胞Ki-67抗原表达降低、Ki-67蛋白降低、凋亡细胞阳性率升高、小鼠肿瘤体积减小。结论:Ki-67基因放射性核素反义治疗能抑制肾癌细胞Ki-67基因表达及细胞增殖、促进凋亡。 Objective: To evaluate the effects of ^125I-labeled antisense oligonucleotide (^125I-A- SODNs) targeting Ki-67 gene on the proliferation and apoptosis of human renal carcinoma cell line in vitro and in vivo. Methods: Antisense oligonucleotides (ASODNs) was coupled with ^125I. Human renal carcinoma cell line 786-0 cells and implanted tumor mice were treated with ^125I-ASODNs. The Ki-67 expression was detected using immunohistochemical technique and Western blot method, respectively. Apoptosis of 786-0 cells was detected by TUNEL assay. Results: The Ki-67 positive expression rate and Ki-67 protein rate were low, but the rate of apoptosis was high. The tumor volume was significantly small. Conclusion: Effects of ASODNs targeting Ki67 gene on the proliferation and apoptosis of human renal carcinoma cells are augmented when ASODNs coupled with ^125I.
