摘要
目的研究莪术油注射液在体外对人子宫内膜癌细胞株RL-95-2细胞周期进程的干扰及其可能的机制。方法通过MTT法实验观察莪术油在体外对RL-95-2的抑制作用;流式细胞仪分析其细胞周期的改变;免疫细胞化学法观察部分细胞周期素的表达。结果莪术油注射液对RL-95-2的抑制作用呈时间-剂量依赖性,中效浓度(72h)为222.36μg/ml,1/2此剂量的莪术油作用于Rl-95-2细胞株24小时、48小时、72小时均可引起G0/G1期细胞比例增加,S期、G2/M期细胞比例减少,统计学表明这种改变差异有非常显著性(P<0.01)。免疫细胞化学法显示Cy-clinD1、CyclinE、P16表达增加,CyclinA、突变型P53表达减低。结论莪术油注射液在体外可抑制人子宫内膜癌细胞株RL-95-2增殖,可能系通过促进P16表达增加,而改变细胞周期进程,同时抑制突变型P53表达从而抑制细胞生长的。
Objective:To investigate the in vitro inhibited cell cycle of the Human endometrial carcinoma cell line RL- 95 - 2 caused by Eleo - Zedoray parenteral solution and its mechanism. Methods: MTT assay were used to display the sensitivity of in vitro RL - 95 - 2 cells inhibited by Eleo - Zedoray parenteral solution. FCM were used to observe the alteration of the RL- 95 - 2's cell cycle. The expressions of Cyclin D1, Cyclin E, Cyclin A, P16, P53 were observed by imrnunocytochemical assay. Results:The antiproliferative effect of Eleo-Zedoray parenteral solution to RL - 95 - 2 is dose - dependent and time- dependent. The half dosage of the IC50(72h) (222.36μg/ml) of Eleo - Zedoray parenteral solution can cause the proportion of G0/G1 phase to raise and at the same time the proportion of S phase, G2/M phase to decrease at 24, 48 and 72 hours. Statistic analysis show significant difference compared with the control group. Immunocytochemical assay displayed that the expression of CyclinD1, Cyclin E, P16 was increased while the expression of Cyclin A, mutant P53 was reduced. Conclusions: Eleo - Zedoray parenteral solution can inhibit the Human Endometrial carcinoma RL- 95 - 2 call line proliferation by promoting the expression of P16 and inhibit the expression of mutant P53.
