摘要
目的探讨干扰素诱导蛋白-10(IP-10)mRNA表达与非特异性角膜炎症反应的相关性及核因子NF-kB抑制剂对IP-10表达的阻断作用。方法建立BALB/C鼠非特异性角膜炎动物模型,对各组角膜进行临床和病理组织学观察,通过RT-PCR对各组角膜不同时相点IP-10的转录水平进行检测。结果角膜缝线及联合结膜下LPS注射均能够诱导角膜IP-10较高水平的表达;IP-10的表达与角膜炎症细胞浸润的加重程度密切相关;IP-10的表达可被NF-kB抑制剂PDTC部分阻断。结论角膜缝线联合结膜下脂多糖(LPS)注射能够诱导明显的角膜非特异性炎症反应;这种炎症反应与IP-10的表达密切相关;NF-kB对IP-10的表达起着重要的调控作用。
Objective To analyze the relationship between interferon-inducible protein10(IP-10) mRNA expression and corneal inflammation and to assess the inhibition of nuclear factor-kB (NF-kB) inhibitor on IP-10 mRNA expression. Methods The models of corneal suture alone or combined with subconjunctival injection were established. Forty-eight BALB/c mice were allocated into suture group, Suture + LPS group, Suture + LPS + PDTC group randomly and other 4 mice were used as control group. Clinic manifestations and pathologic changes of mice corneas in various groups were observed by slit lamp microscope and transmission electron microscopy (TEM) respectively. At postoperative dayl, 3, 7 and day 14, the corneas of each mouse in different group were excised and IP-10 mRNA expression was examined by TEM and reverse polymerase chain reaction (RT- PCR). Results The corneal IP-10 mRNA relative value at 1, 3, 7 days in suture group(0.72±0.08, 0.65±0.15, 0.56±0.09, respectively)and Suture + LPS group (1.10±0.16, 1.03±0.11, 0.94±0.09, respectively) was higher than that of control group(0.27±0.07). On 14 days in postoperation (0.43±0.10), that in Suture + LPS + PDTC group(0.67±0.13, 0.58±0.05, 0.38±0.08 respectively), however, was much lower than that of Suture + LPS group at 1, 3, 7 days, showing a increasing trend in IP-10 mRNA expression with the growth of inflammatory corpuscles in cornea within 7 days after operation and a inhibiting effect of PDTC on IP-10 mRNA. Conclusion The corneal suture combined with LPS subconjunctival injection can induce obvious nonspecific keratitis, which has a close relationship with the expression of IP-10 mRNA. The activity of NF-kB has a great influence on the regulation of IP-10 mRNA expression.
