灯盏花素对肢体缺血再灌注骨骼肌影响的实验研究 被引量:2

摘要 目的:探讨灯盏花素对肢体缺血再灌注骨骼肌的保护作用。方法:健康成年家兔20只,随机分为对照组和实验组,每组10只,建立兔肢体缺血再灌注损伤动物模型。在即将恢复血流灌注时,两组分别自耳缘静脉注射生理盐水(对照组)、灯盏花素注射液(实验组)。分别在缺血前、缺血后2h,再灌注后1h、再灌注后3h采集术侧股静脉血样,测定血清乳酸脱氢酶(LDH)、肌酸激酶(CK)的含量。取上述各时相点动物胫前肌组织,测定骨骼肌组织湿/干重比值并观察肌组织微细和超微结构变化。结果:实验组再灌注后LDH、CK明显低于对照组(P<0.01),骨骼肌组织湿/干重比值低于同期对照组(P<0.05)。光镜及电镜下观察实验组骨骼机损伤轻于对照组。结论:灯盏花素能够缓解组织水肿,保护缺血再灌注骨骼机。 Objective:To study the protective effect and mechanism of Breviscapine on ischemia reperfusion injury of skeletal muscle.Methods: The models of ischemia reperfusion injury in extremities were estabhshed in 20 healthy adult rabbits, then were randomly divided into experimental and control groups. Normal saline injection and Breviscapine were infused intravenously into the control and experimental rabbits respectively just before blood reperfusion. At the points of pre-ischemia,2 hours after ischemia, 1 and 3 hours after repeffusion,the blood samples from the femoral veius in operated side of these rabbits were collected respectively for measurements of lactic dehydrogenase(LDH), creatine kinase(CK) by means of biochemical methods. Meanwhile, the tibialis anterior muscles were harvested for detecting Wet/Dry ratio of skeletal muscle and observing their refined and ultrastructures, Resuits: After reperfusion, the contents of LDH and CK decreased markedly in the experimental group compared with control group( P〈0.01 ). The Wet/Dry ratio of skeletal muscle was lower than concurrent control( P〈0.05). The injuries of skeletal muscles from experimental group were mere hght compared with those from control group by means of hght microscopy and electronic microscopy. Conclusion: Breviscapine could effectively reheve swelling of soft tissue and protect skeletal muscle from ischemia repeffusion injury.
