大鼠寡霉素敏感相关蛋白(OSCP)基因点突变的改造

摘要 目的获取具有正确核苷酸序列的大鼠寡霉素敏感相关蛋白(OSCP)基因。方法通过限制性核酸内切酶将发生有义突变的两个OSCP基因的DNA克隆进行改造,并进行基因重组。结果酶切及测序显示对OSCP基因有义突变部分的纠正获得成功。结论通过基因工程技术获得具有正确核苷酸序列的大鼠寡霉素敏感相关蛋白(OSCP)基因,为OSCP的表达及结构功能研究奠定了基础。 Objective To obtain the correct nucleotide sequence of oligomycin sensitivity-conferring protein (OSCP) of rats. Method The two clones of OSCP gene were modified by restriction endonuclease and recombined by T4DNA ligase. Result Restriction endonuclease digestion and sequencing showed that the modification of the sense mutation of CLSCP gene can be successful. Conclusion The correction nuclcotide sequence of OSCP gene was successfully obtained through gene engineering, providing a basis for the research of OSCP gene expression.
