摘要
目的探讨大鼠脑组织缺血后不同时间胶质原纤维酸性蛋白(glial fibrillary acidic protein,GFAP)的表达,旨为法医实践中推断脑组织缺血时间提供科学的参考依据。方法建立Wistar大鼠脑组织缺血模型,应用免疫组化S-P法与计算机图像分析技术检测脑组织缺血后GFAP在缺血后不同时间以及正常组的表达。结果随着脑组织缺血时间的延长,GFAP在星形胶质细胞的阳性表达逐渐增高,缺血1 h,GFAP的阳性表达与正常组比较无差异;3 h与1 h比较有差异(P<0.05);6-24 h各组间比较有显著性差异(P<0.01)。结论大鼠脑组织缺血GFAP阳性表达呈现一定规律,与缺血时间明显相关,可作为推断较长脑缺血时间的参考指标之一,在法医检案中具有应用价值。
Objective To study the expression of glial fibrillary acidic protein (GFAP) after cerebral ischemia, in order to find out a method of estimating the time of cerebral ischemia in forensic medicine aspect. Methods After setting up an model of experimen- tal focal cerebral ischemia in rats, immunohistochernical method was applied to ol^xve the changes of GFAP in the cerebral and the immunostaining results were assessed by computer imaging analysis system and statistic analysis. Results The expression of GFAP increased following the time of cerebral ischemia. At 1 h after ischemia, GFAP-positive compared with the normal group were not different. The expression of GFAP had become obviously from 1 h to 3 h (P 〈 0.05), and significantly stronger than previous one after 6 h (P 〈 0.01 ). Conclusion Expression of GFAP is correlated with the time of cerebral ischemia in rats, suggesting that the expression of GFAP may be served as the marker for timing of brain ischemia in forensic practice.
