人巨细胞病毒多肽抗原的免疫原性分析

Analyses of Immunogenicity of Human Cytomegalovirus Polypeptide Antigens

目的分析人巨细胞病毒多肽抗原的免疫原性强弱,寻求与早期诊断相关的抗原成分。方法应用SDS-聚丙烯酰胺凝胶电泳和免疫印迹技术对比分析细胞抗原与病毒抗原的差异,确定病毒多肽抗原的存在,利用不同时期免疫动物的多克隆抗体分析病毒多肽抗原的免疫原性。结果SDS-聚丙烯酰胺凝胶电泳和多克隆抗体免疫印迹结果表明分子量为65kD和72kD的人巨细胞病毒多肽抗原免疫原性较强,分子量为51kD和58kD的病毒多肽抗原免疫原性较弱,分子量为150kD的抗原免疫原性最弱。结论65kD和72kD的人巨细胞病毒多肽抗原可以应用于病毒感染的临床确诊。

Objective To analyze the polypeptide antigens immunogenicity of Human cytomegalovirus （HCMV） to find out the antigens with correlation of early diagnosis. Methods Using SDS-PAGE and immunoblots technique to analyze antigens of cell and virus polypeptide antigens to ascertain polypeptide antigens of HCMV. Using the polyclonal antibodys in different periods to analyze the immunogenicity of HCMV polypeptide antigens. Results 65kD and 72 kD HCMV polypeptide antigens were strong immunogenicity and 51kD and 58kD antigens were weak. Conclusion 65kD and 72kD HCMV polypeptide antigens can be used in early diagnosis of HCMV infection.
