摘要
目的探讨化疗药物体外对胆管癌细胞系QBC 939作用的敏感性及对PTEN蛋白表达的影响。方法采用M TT法和流式细胞术检测常用的六种化疗药物体外对胆管癌细胞株QBC 939的敏感性,并运用W est-ern B lot法检测药物体外作用于胆管癌细胞系QBC 939后PTEN蛋白的表达。结果六种化疗药物对于胆管癌细胞的敏感性依次为5-氟脲嘧啶、丝裂霉素、长春新碱、阿霉素、顺铂、氨甲蝶呤。与对照组比较,六种化疗药物体外作用于胆管癌细胞系QBC 939 48h后,PTEN的表达量有不同程度的增高。结论化疗药物体外作用于QBC 939细胞系,可通过上调PTEN的表达最终达到抗癌的作用。
Objective: To investigate the effect of chemotherapeutic drugs on the expression of cholangiocarcinoma QBC939 in vivo and the expression of the tumor suppressor gene PTEN. Methods: By using MTT method, cytofluorometry and Western Blotting assay, the sensititvity of 6 kinds of chemotherapeutic drugs were detected, and PTEN protein expression was examed in cholangiocarcinoma cell line QBC939 after the administration of 5-FU, MMC, DDP, VCR, ADM, MTX. Results: Six kinds of chemotherapeutic drugs' sensitivity was arranged from high to low as following: 5-FU, MMC, VCR, ADM, DDP, MTX. Conclusion: Chemotherapeutic drugs play anti-cancer roles in cholangiocarcinoma cell line QBC939 by significantly upregulating PTEN protein.
