摘要
目的 研究结缔组织生长因子(CTGF)在大鼠肝纤维化形成中的表达及作用机制。方法 雄性SD大鼠32只,体重250~300g,皮下注射四氯化碳(CCl4)制备大鼠肝纤维化模型,分别于注射后1、4、8周处理动物.采用免疫组化、RT—PCR等方法对肝组织中CTGF的表达进行检测。结果 CCl4注射诱导后,大鼠肝组织中CTGF的表达较正常对照明显增强(P〈0.01).且注射1、4、8周组肝组织中CTGF的表达强度呈逐级递增的趋势(P〈0.01,〈0.05)。CTGF主要表达于肝星状细胞胞质中.CTGF mRNA的表达与其蛋白质水平的改变相一致。结论 CTGF作为一种致纤维化因子,其过表达促进肝星状细胞的活化增殖及细胞外基质的形成,从而促进了大鼠肝纤维化的发生、发展。
Objective: To explore the role of connective tissue growth factor (CTGF) in liver fibrogenisis end it's mechanisms. Methods. Liver fibrosis models of rats were made by subcutaneously injecting with CCl4. Thirty two male SD rats (weight 250-300g) were randomly scarified at 1 -4 and 8 weeks after injection of CCl4 respectively, and their liver tissues were used for examining CTGF by immunohistochemistry stain and reverse transcription polymerase chain reaction (RT-PCR). Results: The expression of CTGF in rats after injection with CCl4 were found chiefly in hepatic stellate cells (HSC) and hepatocytes and all significantly higher than those in normal rats (P〈0. 01). Moreover, the expression of CTGF presents a progressive tendency respectively at 1st, 4th and 8th week after injection with CCl4(P〈0. 01, 〈0. 05). And the expression of CTGF mRNA changed in similar way as that of CTGF protein. Conclusions: The overexpression of CTGF enhances HSC proliferation of rats, and maybe play an important role in liver fibrogenesis of rat.
