摘要 目的建立一种新的复合式老年性痴呆(AD)大鼠模型,用于AD及其治疗药物的研究。方法选用雄性正常老年Wistar大鼠,左侧侧脑室一次性注射5μL聚集态的Aβ25-35,制备AD动物模型。通过水迷宫试验、脑组织Aβ免疫组化染色和银染色,比较AD模型大鼠和正常老年大鼠的差异。结果模型组大鼠的逃避潜伏期明显延长(P<0.01);免疫组化结果示模型组海马、额叶和颞叶Aβ阳性细胞数增加,平均灰度值显著降低(P<0.01),且染出了脑淀粉样血管病变(CAA)、老年斑(SP)和小胶质细胞;模型组Bielschowsky染色可见星形胶质细胞,但并未见到典型的NFTs。结论老年大鼠加左侧侧脑室一次性注射聚集态的Aβ25-35所致的AD模型在一定程度上模拟了AD的发病特点,可以作为AD及其治疗药物研究的一种新模型。 Objective To establish a novel complex rat model of Aged Dementia(AD) for research of AD and its treatment. Methods The male normal old Wistar rat were selected and 5μL accumulation Aβ25-35 was injected into the left lateral ventricle of rat to establish AD animal model. The pathological observation was made by immunohistochemistry. Results The model group showed that the escape latency time increased ( P 〈 0.01 ). Furthermore, the result of immunohistochemistry revealed that the number of Aβ masculine cells in the hippocampus, frontal lobe and temporal lobe in model group increased and the grey scale score decreased(P 〈 0.01 ), CAA, senile plaques and microglias were observed in AD model rat. Astrocytes in Bielschowsky dyeing was found in model group, but typical NFTs was not loud. Conclusion The rat model of AD might be induced by accumulative β-amyloid(25- 35) injected into left lateral ventricle of old rat. This AD model might be used as a novel model for study AD and its drug treatment.
