流式微珠阵列法检测IFN-γ诱导的Jurkat细胞Th1/Th2细胞因子表达 被引量:2

摘要 本研究旨在探讨流式微珠阵列法检测Th1/Th2细胞因子表达及IFN-γ应用于T淋巴细胞白血病治疗的临床价值。以不同浓度IFN-γ诱导培养Jurkat细胞48小时,用流式微珠阵列法检测培养上清液中IL-2、IL-6、TNF-α和IFN-γ表达,并采用流式细胞术检测膜上IL-2受体(CD25)的表达。结果表明:IFN-γ诱导Jurkat细胞IL-2和TNF-α的表达呈浓度依赖性升高,未检测到IL-6表达,CD25表达明显升高。结论:Jurkat细胞能被IFN-γ诱导高表达Th1细胞因子和CD25,流式微珠阵列法能准确客观地反映Th1/Th2细胞因子表达的动态变化。 In order to explore the value of T-interferon (IFN-γ) in therapy of T lymphoid leukemia and role of cytometric bead array (CBA) method in detection of Th1/Th2 cytokines expression, the Jurkat cells were cultured for 48 hours in different concentration of IFN-γ, the expressions of IL-2, IL-6, INF-γ and TNF-α in culture supernatants were assayed by CBA method; CD25 expression was assayed by flow cytometry. The results showed that the expressions of IL-2, TNF-α were enhanced in INF-γ dose-dependent way, and the expression of CD25 was also enhanced, but there was no expression of IL-6. It is concluded that Jurkat cells induced by IFN-γ were able to express high-level of Thl cytokine and IL-2 membrane receptor ( CD25 ), and the CBA method can be used to exactly evaluate the dynamic change of Th1/Th2 cytokines.
