摘要
目的研究口服大剂量维生素C在健康人体内的药动学。方法10名健康志愿者单、多次口服1 g维生素C,采用HPLC法测定血浆中维生素C浓度,计算药动学参数。结果单次口服1 g维生素C,主要药动学参数t1/2为(12.1±2.71)h;ρmax为(22.0±3.18)mg.L-1;tmax为(2.0±0.4)h;AUC0→36 h为(223.9±35.7)mg.h.L-1;AUC0→∞为(254.3±53.3)mg.h.L-1。多次口服维生素C,主要药动学参数ρmax为(26.6±3.50)mg.L-1;tmax为(1.8±0.4)h;ρmin为(8.25±0.90)mg.L-1;ρav为(12.8±0.92)mg.L-1;AUCss为(299.2±23.3)mg.h.L-1;DF为(1.46±0.28)。结论每日口服1 g维生素C,能够达到临床治疗目的。
AIM To study the pharmacokinetics of high-dose Vitamin C acid in 10 Chinese healthy volunteers after oral administration 1 g ascorbic acid. METHODS Vitamin C concentration in plasma was measured by an improved HPLC methods, and the pharmacokinetic parameters were calculated. RESULTS After single dose administration of 1 g Vitamin C, the main pharmacokinetic parameter t1/2, ρmax,tmax AUC0→36 h, AUCo→∞was (12.1 ± 2.71) h, (22.0 ± 3.18) mg.L^-1,(2.0±0.4)h, (223.9±35.7) mg.h.L^-1, (254.3±53.3) mg.h.L^-1, and that ρmax,tmax,ρmin ,ρav,AUCss,DF was (26.6± 3.50) mg.L^-1,(1.8 ± 0.4)h,(8.25 ±0.90) mg.L^-1, (12.8±0.92) mg.L^-1, (299.2 ± 23.3) mg.h.L^-1,1.46±0.28, after multiple-dose administration, respectively. CONCLUSION In clinic, the goal of treatment was achieved at a dose of 1 g daily.
