摘要 目的研究胰腺癌组织APE的表达及其与临床肿瘤指标的关系。方法(1)通过逆转录-聚合酶链反应(RT-PCR)方法检测4株胰腺癌细胞株APEmRNA的表达;(2)收集37例手术切除的胰腺癌及12例相应癌旁胰腺组织石蜡标本,应用EnVision免疫组化法分别检测癌组织和癌旁胰腺组织APE的表达情况。结果4株胰腺癌细胞株均有APEmRNA表达;37例胰腺癌中APE阳性表达33例(89.2%),其中APE胞核阳性表达7例(21.2%),胞质阳性表达5例(15.2%),胞质及胞核均表达21例(63.6%)。12例癌旁胰腺组织导管及腺泡呈阴性表达,胰岛胞质呈阳性表达,胰腺上皮内瘤变呈胞核阳性表达。APE阳性率与肿瘤分化程度,肿瘤大小、淋巴及远处转移无关(P>0.1)。结论APE在胰腺癌组织中有较高的阳性表达率,检测其定位的特异性表达可能有助于胰腺癌的早期诊断。 Objective To detect expression of APE in human pancreatic carcinoma and discuss its relationship to differentiation degree, size, invasion and metastasis of human pancreatic cancinoma. Methods APE mRNA expression in pancreatic cancer cell line was detected using reversetranscription PCR (RT-PCR), and expression of APE/ref-1 in pancreatic cancer tissue and adjacent normal pancreatic tissue was detected by immunohistochemical EnVision method. Results APE mRNA expression was confirmed by RT-PCR in 4 pancreatic cancer cell lines; the positive expression rate of APE was 89.2% (33/37) in pancreatic cancer tissue, of which 21.2% (7/33) was expressed in APE nuclei only, 15. 2%(5/33) in cytoplasm only, and 63. 6%(21/33) in nuclei and cytoplasm simultaneously. APE expressed in cytoplasm of pancreatic islet cells beside tumor in 6 cases. In pancreatic carcinoma most APE expressed in nuclei and cytoplasm, and beside the tumor. APE was positive in cytoplasm of pancreatic islet cells hut negative in pancreatic duct cells and acini. The positive rate of APE was associated with differentiation, size and lymph node or distant metastasis of the tumor. Conclusions Detection of APE expression has diagnostic implication, and strong APE expression in pancreatic cancer might be used as a parameter for determining the malignancy degree of a tumor.
