摘要
目的探讨S100A4基因mRNA表达与肾癌细胞分化、转移的关系。方法提取31对肾癌及正常组织配对标本的总RNA,用RT-PCR方法测定S100A4基因的表达,并结合临床资料,对S100A4基因差异表达与肾癌病人临床表现的相关性进行研究分析。结果29例标本肿瘤组织中S100A4基因mRNA表达明显高于癌旁正常肾组织;其中低分化肿瘤组病人的阳性表达率高于高分化组(7.94vs5.06,P<0.001);发生转移(包括淋巴结转移和远处转移)者高于无转移者(9.61vs5.53,P<0.001);低年龄组病人(≤50岁)阳性表达率与高年龄组(>50岁)差异无显著性(6.31vs6.66,P>0.05);肾颗粒细胞癌组与透明细胞癌组差异无显著性(6.98vs6.02,P>0.05);肿瘤直径≤5cm组与>5cm组差异无显著性(5.95vs6.93,P>0.05)。结论S100A4基因表达与肾细胞癌分化有关,癌组织S100A4mRNA测定有助于判定肿瘤的预后;S100A4基因差异表达与肾细胞癌转移明显相关,肾癌组织中S100A4基因mRNA可望作为肾细胞癌的病情发展及指导临床治疗的标记物之一。
[Objective] To study the relationship between S100A4 mRNA expression level and differentiation and metastasis of renal cell carcinoma combined clinical data and pathology together. [Methods] This study was based on tissues from 31 patients with renal cell carcinoma treated surgically from 2001 to 2004. After RT-PCR amplification on 31 RCC tissues and paired nom^al kidney tissues besides the tumor, we detected fluorescence dosage of S100A4 mRNA in tissues, and we obtained S100A4 mRNA gene fragments(212 bp). [Results] S100A4mRNA expression was detected in all of the RCC tissues and para-carcinoma tissues using fluorescence. Expression of 29 RCC tissues' S100A4 mRNA was higher than paired para-carcinoma tissues. Of all cases, patients of low-differentiation RCC had higher expression rate than high-differentiation RCC obviously(7.94 vs 5.06,P 〈0.001 ); patients of metastasis also had higher expression rate than non-metastasis(9.61 vs 5.53, P 〈0.001 ). There was no clearly difference of expression rate between Granular cell carcinoma and Clear cell carcinoma (6.98 vs 6.02,P 〉0.05). [Conclusions] S100A4 mRNA is correlated to RCC differentiation and may play an important role in prognosis of RCC. Differential expression of S100A4 mRNA is correlated to metastasis of RCC. S100A4 may be one of markers on assessment of patient's condition developing and clinical treatment instruction in RCC.
