一株伤寒沙门菌检出TEM-1耐药基因

摘要 目的:研究水产品中分离的一株伤寒沙门菌NB2005220中检出的TEM-1耐药基因,分析其在细菌耐β-内酰胺类抗生素中的作用。方法:纸片扩散法药物敏感实验检测细菌耐β-内酰胺类抗生素情况,用常规PCR方法扩增TEM-1耐药基因,并对PCR产物进行测序和NCB I比对分析。结果:发现伤寒沙门菌NB2005220携带TEM-1耐药基因,测序分析其与AJ634602有高度同源性,药敏实验表明,该菌株对氨苄青霉素和羧苄青霉素耐药,对头孢噻吩等敏感,三相水解实验证明这种耐药为β-内酰胺酶引起。结论:首次从水产品中分离的沙门菌NB2005220携带TEM-1耐药基因,介导该菌对氨苄青霉素、羧苄青霉素等耐药。 Objective:To study the TEM - 1 drug resistance gene detected in strian NB2005220 of Salmonella enterica serovars Typhimurium( S. Typhimurium) isolated from the sea food and explore the function of the TEM - 1 in resistance against β -lactam antibiotics. Methods:Susceptibility to β- lactam antiobiotics was tested with plate diffusion method. Routine PCR was performed to amplify the TEM - 1 gene and PCR product was sequenced and blasted via NCBI Blast. Results :TEM - 1 resistance gene was identified in strain NB2005220 and was found to be highly homologous to AJ634602 through sequencing. The isolate was resistant against ampicillin and carboxypeniciUin. Three - dimensional test was applied to prove that β - lactamase caused this resistance. Conclusion:TEM -1 drug resistance gene is,for the first time, first detected in & Typhimurium from sea food which might cause the resistance against ampicillin and carboxypenicillin.
