蟾酥中吲哚烷胺类生物碱的分离与结构鉴定 被引量:13

摘要 目的对蟾酥(Venenum Bufonis)中吲哚烷胺类生物碱进行提取分离和结构鉴定。方法蟾酥水溶性部分经HP 20树脂柱,制备薄层色谱,高效液相色谱分离和精制,得到6个吲哚烷胺生物碱,通过理化常数和光谱数据,分析鉴定了其化学结构。结果蟾酥水提取物中分离得到6个生物碱,分别鉴定为蟾毒色胺(bufotenine,Ⅰ)、蟾毒色胺内盐(bufotenidine,Ⅱ)b、ufobutanoic acid(Ⅲ)、5羟色胺(serotonin,Ⅳ)b、ufotenine N oxide(Ⅴ)、N甲基5羟色胺(Ⅵ)。结论化合物Ⅴ和Ⅵ为首次从蟾酥中分离得到。首次对蟾酥色胺内盐及bufotenine N oxide的1H1、3C NMR数据进行归属。 Objective To extract and isolate the indolealkylamines from the traditional Chinese medicine Toad Venom and to elucidate their structures. Methods The indolealkylamines were extracted and isolated by macroporous resin HP-20, finally, it was separated by preparative TLC and semipreparative HPLC with an ODS column. Their structures were identified through the studies of physico-chemical properties and spectroscopic analysis. Results The compounds obtained were identified as bufotenine( Ⅰ ), bufotenidine( Ⅱ ), bufobutanoic acid ( Ⅲ ), serotonin (Ⅳ ), bufotenine N-oxide (Ⅴ ) and N-methyl serotonin ( Ⅵ ). Conclusions Compound Ⅴ and Ⅵ are isolated from Toad Venom for the first time.
