除草剂百草枯对大鼠胎儿动脉管的毒性作用

Toxic effects of bipyridine herbicide paraquat on ductus arteriosus of rat fetus

目的研究除草剂百草枯对胎儿动脉管的毒性影响。方法(1)取妊娠21 d大鼠100只,随机分成每组20只。皮下注射25 mg/kg百草枯,于不同时间测定母体和胎儿血液中百草枯浓度;(2)取妊娠21 d大鼠随机分成4组,实验组分别皮下注射2、7和25 mg/kg百草枯后,于不同时间观察胎儿动脉管收缩情况;(3)取妊娠19 d(上午)1、9 d(下午)、20和21 d的大鼠各8只,随机分成2组。实验组于不同时间皮下注射25 mg/kg百草枯,3 h后观察动脉管收缩情况。结果百草枯在母体及胎儿血液中呈不同迁移。百草枯可引起胎儿动脉管收缩,且存在剂量-效应关系。百草枯引起胎儿动脉管收缩的临界期在胎龄19.5和19.7 d之间。结论百草枯对胎儿动脉管具有毒性作用。

Objective To study the toxic effects of paraquat on ductas arteriosus of rat fetus.Methods （1） 100 gravid female wistar rats on 21 days of gestation were respectively divided randomly into 20 rats per group.The rats were administered with paraquat by subcutaneous injection at the dose of 25 mg/kg. The concentration of paraquat in plasma of mother＇ body and the fetuses were determined. （2） The gravid female wistar rats on 21 days of gestation were respectively divided randomly into 4 groups. In the experimental group, the rats were administered with paraquat by subcutaneous injection at a dose of 2,7,25 mg/kg. Fetuses were used for observing contraction of ductus arteriosus per gravid rat on different times after injection. （3）Eight gravid female wistar rats on day 19, 19,20,21 of gestation were respectively divided randomly into 2 groups. In the experimental group, the rats were administered with paraquat by subcutaneous injection at the dose of 25 mg/kg respectively on different times. The fetuses were gotten 3 hours after injection for observing contraction of ductus arteriosus per gravid rat. Results Paraquat showed different migration in plasma of mother＇ body and fetus. Paraquat can induce contraction of ductus arteriosus of rat fetus. The critical contraction time of ductus arteriosus of rat＇ s fetus induced by paraquat is between 19.5 days and 19.7 days. Conclusion Puraquat exerts toxicity effect on ductus arteriosus of rat fetus.
