摘要
[目的]比较双歧杆菌及其发酵液的免疫及抗肿瘤作用,探讨其抑瘤作用和相关机制。[方法]观察两种肿瘤细胞荷瘤小鼠的生存期。右腋下荷瘤后,进行病理切片观察。MTT法计算给予不同方法后的抑瘤率,并计算淋巴细胞转化率。流式细胞仪和TUNEL技术检测凋亡情况。制备电镜标本,观察其超微结构。[结果]双歧杆菌能够延长荷瘤小鼠的生存期,病理切片可见大量炎性细胞浸润。流式细胞仪检测发现,死菌液、活菌液作用后肿瘤细胞凋亡率分别(11.36±3.20)%和(8.08±2.00)%为均高于对照组(1.80±0.50)%(P<0.01),TUNEL技术检测,对照组髓过氧化物酶(MPO)染色阳性率(0.8±0.3)%显著低于活菌液组(1.2±0.4)%(P<0.05)。[结论]双歧杆菌死菌液、活菌液在体内外均有较好的抑瘤作用,发酵液并未显示出抑瘤作用,说明双歧杆菌的抗肿瘤作用主要来源于菌体的作用。
[Objective] By comparing Bifidobacterium and its broth's immunization and anti- tumor effect, we initially understand its anti - tumor mechanism. [ Method] BABL/C mice bearing H22 and S180 were given to different methods. The therapeutic effect was determined by life span. Otherwise, the therapeutic effect was determined by means of pathological examination of tumor tissue. MTT assay calculats the Inhibition rate, and calculats lymphocyte ransformation rate. Apoptosis were detected by Flow cytometry and the transferase- mediated dUTP nick end labeling (TUNEL) technique. Electron microscope was used to study the cell morphology. [ Result] Thanato- Bifidobacterium and vivi- Bifidobacterium could prolong the life span of mice bearing tumor, inhibited growth of tumor. The apoptosis rate was (11.36 ± 3.20)% and (8.08 ± 2.00)% respectively, both higher than that of the control group ( 1.80 ± 0. 50) % , ( P 〈 0. O1 ). The positive rate of H22 by M PO staining was lower in the control group (0.8 ± 0. 3) % than the vivi - Bifidobacterium group(1.2 ± 0. 4)% (P 〈 0.05). [Conclusion] Thanato- Bifidobacterium and vivi- Bifidobacterium have signifycant anti- tumor effect in vitro and in vivo, could prolong the life span of mice bearing tumors and inhibit substantial tumor ceils , but its broth did not show anti- tumor effect , demonstrate that the anti- tumor effect of Bifidobacterium chiefly origins from its thalline.
