电针对复合应激致慢性疲劳大鼠下丘脑组织中单胺类神经递质含量的影响 被引量:29

Effects of Electroacupuncture on Contents of Monoamines Transmitters in Hypothalamic Tissues of Chronic Fatigue Syndrome Rats

摘要 目的:观察电针“百会”“太溪”对复合应激致慢性疲劳大鼠脑组织中单胺类神经递质含量及行为学的影响。方法:选取二级Wistar雄性大鼠36只,随机分为正常组、模型组和电针组,采用慢性复合应激法建立慢性疲劳大鼠模型,电针“百会”“太溪”,测定大鼠行为学、下丘脑组织中单胺类神经递质含量的变化及针灸的治疗作用。结果:模型组下丘脑组织中单胺类神经递质含量降低,大鼠行为学的改变支持慢性疲劳的情况;电针治疗可使以上指标趋于正常。结论:复合应激致慢性疲劳大鼠脑组织中单胺类神经递质含量减少可能参与复合应激所致的慢性疲劳的发生与发展,电针治疗有良好的作用。 Objective: To observe the effect of electroaoupuncture (EA) of "Baihui" (GV 20) and "Taixi" (KI 3) on changes of behavior activities and concentrations of monoamines transmitters in hypothalarnic tissues of rats with chronic fatigue syndrome (CFS). Methods: Thirty-six male Wistar rats were evenly randomized into control, model and EA groups. CFS model was established by intramuscular injection of hydrocortisorle (20 mg/kg, 1 time/d, 9 d) and withdrawal of hydrocortisone, and forced swim in cool water (30 min/1 time/d, 21 days). EA (2/20 Hz, 9 V) was applied to "Baihui" (GV 20) and "Taixi" (KI 3) for 30 min, once daily and continuously for 21 days. Open-field test, rat's tail suspension immobility test and exhausted swimming test were used to check the rats' behavioral changes and double antibody sandwich enzyme-linked imrnunceorbnent assay (ABCELISA) was used to determine concentrations of noradrenaline (NE), serotonin (5-HT) and dopamine (DA) in hypothalamus tissues. Results: Compared with control group, the crossing and rearing activity times, the duration of tail-suspension immobility arid the duration of exhausted swim, and the contents of NE, 5-HT and DA of hypothalamus tissues in model group were all significantly lower (P〈0.05); while the values of the abovernentioned 7 indexes of EA group were all significantly higher than those of model group ( P 〈 0.05). No significant differences were found between EA group and control group in these 7 indexes ( P 〉 0.05). It showed that EA could improve behavioral activities of CFS rats and raise the contents of monoamines neurotransmitters in the hypothalamus. Conclusion: EA of "Baihui" (GV 20) and "Taixi" (KI 3) can ameliorate behavior activities of CFS rats and reverse the declined contents of NE, 5-HT and DA of hypothalarnus tissue.
