补阳还五汤及其有效部位生物碱、苷对大鼠动脉血栓形成纤溶相关指标的影响 被引量:12

Effect of Buyang Huanwu Decoction and Its Active Fractions Alkaloid and Glycoside on Fibrinolytic System in Rat Model of Arterial Thrombosis

摘要 目的研究补阳还五汤及其生物碱、苷两类有效部位对大鼠颈总动脉血栓形成纤溶相关指标的影响。方法将SD大鼠84只随机分为补阳还五汤原方组、生物碱组等7组。先给大鼠喂药后以FeCl3诱导左侧颈总动脉血栓形成,以发色底物法测定血浆组织型纤溶酶原激活物(tPA)、纤溶酶原激活物抑制物(PAI)和纤溶酶原(PLG)活性,并测定血栓质量。结果①FeCl3可致颈总动脉血栓形成。补阳还五汤、生物碱、苷、抵克利得和低分子肝素均可使血栓质量减轻(P<0.05)。②模型组tPA和PLG活性均降低(P<0.05),PAI活性无显著变化(P>0.05)。与模型组比较,补阳还五汤原方可使血浆tPA、PLG活性升高(P<0.05),而生物碱和苷对tPA和PLG活性无影响。结论低分子肝素(LMWH)和抵克利得可抑制由FeCl3诱导的大鼠血栓形成,表明该模型与凝血功能亢进和血小板活化有关。补阳还五汤及其有效部位生物碱和苷也可抑制该模型血栓形成,原方可使血浆tPA活性和PLG活性升高,提示其作用与促进纤溶功能有关;而生物碱和苷对tPA、PAI和PLG无明显影响,表明其抗血栓作用不是通过促进纤溶而实现的,其作用可能与其它环节有关。 Objective To investigate the effect of Buyang Huanwu Decoction (BHD) and its active fractions such as alkaloid and glycoside on carotid arterial thrombosis in rats. Methods The rats, divided into 7 groups, were administered with test drug, then the left carotid artery appled FeCl3 to form thrombus models. The activities of tissue plasminogen activator (tPA), plasminogen activator inhibitor (PAI) and plasminogen(PLG) in plasma were determined by chromogenic substrate assay, and thrombi weight was measured. Results ①Formation of thrombus could be induced by FeCl3 in carotid artery. Weight of thrombi reduced (P〈 0.05) by BHD, alkaloid, glycoside, LMWH and ticlopidine. ②The activities of tPA and PLG in the model group decreased (P〈0.05), the activities of PAI had no significant change. Compared with the model group, the activities of tPA and PLG in BHD was increased (P〈0.05), but the activities of tPA and PLG in alkaloid and glycoside group were not affected (P〉0.05).Conclusions BHD and its active fraction can inhibit thrombosis in the model. BHD can enhance the activities of tPA and PLG, which suggests that effect of BHD may be associated with accelerating fibrinolysis. Alkaloid and glycoside are not effective on the activities of tPA, PAI and PLG, which means that antithrombotic action of alkaloid and glycoside is not based on accelerating fibrinolysis, may be related to other aspects.
