菹草类胡萝卜素抗癌作用与机理的研究 被引量:5

摘要 通过MTT细胞增殖抑制试验,测定肿瘤细胞膜唾液酸(SA)含量、线粒体跨膜电位(ΔΨm)、细胞培养液中NO和NOS含量等的变化,研究菹草类胡萝卜素(carotenoids extracts fromPotamogeton crispusL.,CEPC)对人宫颈癌Hela细胞株增殖的影响及其作用机制。结果表明:CEPC对Hela细胞增殖有明显的抑制作用;能显著降低Hela细胞表面的SA含量,诱导Hela细胞ΔΨm下降,导致iNOS活性增强,NO含量显著升高。CEPC对Hela细胞的生长抑制作用可能是通过降低肿瘤细胞膜SA含量,使线粒体跨膜电位下降i、NOS的活性增强、NO的浓度升高而引起肿瘤细胞凋亡。 The cell growth inhibition was tested in MTT assay. The sialic acid (SA) content in Hela cell membrane, mitochondrial membrane potential(△ψm ) and NO content, NOS activity in culture supernatant were measured to explore the antitumor mechanism of carotenoids extracted from Potamogoton crispus L. (CEPC). Results showed that CEPC had strong inhibitive effects on the cell growth and reduced the SA content in cell membrane and △ψm obviously, iNOS activity and NO content were significantly improved by CEPC. CEPC can inhibit Hela cell proliferation and the antitumor mechanism maybe reduce SA content in cell membrane,induced cell apoptosis by △ψm reduction and increase of iNOS activity and NO content.
