摘要
目的经β淀粉样蛋白(Aβ)作用后海马神经细胞的死亡方式及其意义。方法观察不同浓度(分为浓度效应组和浓度效应对照组)、不同时间(分为时间效应组和时间效应对照组)Aβ1-40对体外培养的海马神经细胞的毒性,并用透射电镜、琼脂糖凝胶电泳、荧光染色进行细胞死亡的检测。结果经51、0、20μmol/L Aβ1-40处理12 h后,神经细胞凋亡和胀亡率明显高于浓度效应对照组;给予10μmol/L Aβ1-40处理的条件下,经过61、2、24 h后,神经细胞凋亡和胀亡率明显高于时间效应对照组。结论一定浓度的Aβ1-40对体外培养的海马神经细胞具有细胞毒性,并且诱导神经细胞死亡具有时间和剂量相关效应。
Objective To investigate the modes of hippocampal neurons'death induced by β-amyloid protein (Aβ) .Methods The toxicity of Aβ1-40 to neurons was observed at different concentrations for different timeperiods in vitro. Neuron death was analyzed by electron microscopy, DNA agarose gel electrophoresis and fluorescence staining. Results At 12 h after treating with 5,10,20μmol/L Aβ1-40, apoptosis and oncosis rates of neurons were significantly higher than those of the control group. At 6 h, 12 h and 24 h following adding 10 μmol/L Aβ1-40 to the culture medium, apoptosis and oncosis rates of neurons were significantly higher than those of the control group. Conclusion Certain concentrations of Aβ1-40 showed cytotoxic effect on cultured neurons and induced the neuron necrosis in both dose- and time-dependent manners.
