反相高效液相色谱法测定丹参滴注液中丹参素和原儿茶醛 被引量:10

摘要 目的建立同时测定丹参滴注液中丹参素和原儿茶醛含量的方法。方法利用RP-HPLC法,采用Diamonsil^TM C18色谱柱(250mm×4.6mm,5μm),以甲醇-乙腈-冰醋酸-水(10:5:0.25:85)为流动相,流速为1.0mL·min^-1,检测波长为281nm。结果用RP-HPLC法测定2个活性成分丹参素和原儿茶醛分别在20.6~82.4μg·mL^-1和7.8~56.0μg·mL^-1与峰面积呈良好线性关系,相关系数分别为0.9990和0.9996。回收率丹参素为99.46%(RSD=1.0%,n=3)、原儿茶醛为99.96%(RSD=0.97%,n=3)。结论本方法简便、快速,两峰分离良好,可用于丹参滴注液的质量控制。 OBJECTIVE To determine the contents and determination method of protocatechuic aldehyde (PCAD) and danshensu in danshen dripping solution. METItODS RP-HPLC method was used. The column was Diamonsil^TM C18 (250 mm×4.6 mm, 5μm). The mobile phase was methanol-acetonitrile-acetic acid glacial-water (10 : 5 : 0. 25 : 85) ; the flow rate was 1.0 mL · min^-1 ; the detection wavelength was 281 nm. RESULTS The calibration curves of danshensu and PCAD were linear in the range of 20.6-82.4μg · mL^-1 and 7.8-56. 0μg · mL^-1 with the correlation coefficient beyond 0. 999 0 and 0. 999 6 respectively. The mean recoveries of the assay were 99.46% (RSD= 1.0%, n=3) for danshensu and 99.96 % (RSD=0.97%, n=3) for PCAD respectively. CONCLUSIONS This method is simple, quick with good separation, and it is useful in the quality control of danshen dripping solution.
