摘要
目的探讨氯化镧对人脑胶质瘤SHG44细胞增殖的影响及其作用机制。方法体外培养人胶质瘤细胞,MTT法测定氯化镧对SHG44细胞的增殖影响,电镜检查细胞超微结构的改变,原位杂交技术及免疫组织化学染色技术检测PTEN基因及其蛋白表达情况。结果氯化镧可抑制人脑胶质瘤SHG44细胞的生长,电镜下见胶质瘤细胞核染色质浓缩、核固缩、核溶解,出现凋亡细胞。PTEN基因在蛋白、mRNA水平均显著增强。结论氯化镧可抑制人脑胶质瘤SHG44细胞的生长,并促进凋亡。
Objective To investigate the mechanism of inhibition of LaCl3 on human ghoma SHG44.Metheds The human ghoma SHG44 cells were cultured in vitro, The inhibition effect of LaCl3 on SHG44 cells proliferation was determined by MTT method. The cell ultrastructure were detected by electron microscope. The protein expression and translation of pten gene were detected by immunohistochemistry and in situ hybridization methods. Results The LaCl3 could inhibit the growth of SHG44 and induce apoptosis. The nuclei pycrosized anddissolved, chromatin condensation and aggregation were observed and typical apoptosis cells appeared. The PTEN protein expression and mRNA translation were increased. Conclusion LaCl3 can inhibit the proliferation of SHG44 cells in vitro and cause apoptosis.
