表皮干细胞体外诱导转分化为角膜上皮细胞的实验研究

Transdifferentiation of epidermal stem cells into corneal epithelial cells in vitro

目的探讨表皮干细胞向角膜上皮细胞转分化的可塑性。方法用Ⅳ型胶原筛选的方法培养并鉴定表皮干细胞,经体外与角膜缘基质细胞共培养诱导分化,免疫组化检测角膜上皮细胞特异标志物K12表达。结果获得的表皮干细胞表现出很强的增殖潜能,光镜下为原始细胞形态特征,能强表达K19和β1-integrin,共培养2周后可见细胞分化,细胞表达角膜上皮特征性K12。结论在本实验的体外诱导条件下,表皮干细胞能转分化为角膜上皮细胞。

Objective To explore the plasticity of transdifferentiation of epidermal stem cells （ESCs） into corneal epithelial cells. Methods Epidermal stem cells enriched by adhering to type Ⅳ collage were induced in vitro by cocuhure with corneal stromal fibroblasts. The method of HCC were used to show the expression of K12 which is expressed specially in corneal epithelial cells. Results ESCs with greater proliferation potential strongly expressed K19 and β1- integrin. After two-week induction, the K12 positive cells could be detected. Conclusion The data suggested that the epidermal stem cells have the potential to transdifferentiate into corneal epithelial cells in vitro,
