摘要 目的应用昆明小鼠股骨骨髓嗜多染红细胞微核实验,研究蝙蝠葛酚性碱(phenolic alkaloids from menispermum dauricum,PAMD)的致突变作用和抗突变作用.方法均在10~40 mg/kg剂量下,连续腹腔注射给药11d,在致突变实验,末次给药6 h后处死小鼠,检测骨髓嗜多染红细胞微核率.在抗突变实验,小鼠在于处死前的24 h,腹腔注射40 mg/kg环磷酰胺(cyclophosphamide,CP)或2 mg/kg丝裂霉素C(mitomycinc,MC),末次给药6 h后取样.结果蝙蝠葛酚性碱与对照组相比未诱发骨髓嗜多染红细胞微核率的增高(微核细胞率<3‰).蝙蝠葛酚性碱可使环磷酰胺和丝裂霉素C诱发的骨髓细胞微核率增高数值出现明显的降低.PAMD+CP组微核细胞抑制率分别为61.83%、43.51%和22.52%.PAMD+MC组微核细胞抑制率分别为31.09%、21.50%和8.29%.结论蝙蝠葛酚性碱具有一定的抗突变作用,而无致突变作用. Objective To investigate the antimutagenic and mutagenic activity of phenolic alkaloids from menispermum dauricum (PAMD) by mouse micronucleus test of polychromatic erythrocytes in bone marrow. Methods PAMD at the dose of 10, 20, 40 mg/kg was injected intraperitoneally to mice once a day for 11 d, the physiological saline containing 10% ethanol as normal control, cyclophosphamide (CP) and mitomycin C (MC) as positive controls. In the mutagenic test, the mice were killed at 6 h after the last administration, and the micronucleated polychromatic erythrocytes of bone marrow were observed and measured ; in the antimutagenic test, CP of 40 mg/kg or MC of 2 mg/kg were injected intraperitoneally at 24 h before killing, and the statistic was obtained at 6 h after the last administration. Results The rate of the micronucleated polychromatic erythrocytes ( 〈 3‰) in PAMD group were not significantly different from the control group. PAMD significantly decreased the rate of the micronucleated polychromatic erythrocytes in mice induced by CP and MC as compared with the positive control groups. The micronucleus cell inhibiting rate of PAMD + CP group were 61.83%, 43.51% and 22.52%, and that of PAMD + MC group were 31.09%, 21.50% and 8.29%. Conclusion Phenolic alkaloids from menispermum dauricum have antimutagenic effect. The mutagenicity of phenolic alkaloids from menispermum dauricum was not observed in our study.
