摘要 目的建立大鼠急性乙醇中毒昏迷模型,观察orexinA对昏迷动物的促醒作用。方法取成年雌性SD大鼠74只,分别通过腹腔注射乙醇的方法建立大鼠急性乙醇中毒昏迷实验模型。通过翻正反射判断大鼠意识状态的改变;通过脑电图判断脑电活动变化。结果给予腹腔注射32%乙醇(14ml/kg)后,大鼠行为状态和脑电图变化符合昏迷诊断标准。同对照组比较,高剂量orexinA治疗组(7.70nmol/kg)大鼠翻正反射消失持续时间明显缩短,行为觉醒改变较为显著,而低剂量治疗组(3.85nmol/kg)行为变化不明显。此外,给予高剂量orexinA可引起大鼠脑电活动改变,同给药前比较,δ波显著减少。结论侧脑室使用orexinA对昏迷大鼠具有促醒治疗作用。 Objective To establish a rat model of acute alcohol intoxication-induced coma and to investigate roles ot orexin-A in treatment of coma. Methods coma in 74 adult male Sprague-Dawley rats. Changes in flex (LOR). Electroencephalogram (EEG) recordings Intraperitoneal injection of alcohol was used to cause the conscious state were judged by loss of righting rewere used to evaluate the changes in activity of rat brain. Results After administration of 32% alcohol ( 14 ml/kg), rats met the diagnosis criterion of coma. In behavioral observations, both behavioural states and EEG patterns of orexin-A at dose of 7.70 nmol/kg induced significant decrease in the duration of loss of righting reflex (LOR) as compared with the control group, whereas orexin-A at dose of 3.85 nmol/kg had no obvious effects on the duration of LOR. In EEG experiments, 7.70 nmol/kg orexin-A markedly reduced δ activities in coma rats. Conclusion The intracerebroventricularl injection of orexin-A has a potent arousal effect on acute alcohol intoxication-induced coma of rats.
