Determination of the Organic and Inorganic Selenium Contents in Selenium-Enriched Yeast by Hydride Generation-Atomic Fluorescence Spectrometry

摘要 建立了氢化物发生-原子荧光光谱法测定富硒酵母中硒的分析方法。研究了仪器的工作条件、试剂对硒测定的影响;探讨了消化试剂对样品消化的影响和共存离子的干扰及消除。在优化的工作条件下,硒的检出限为0.078μg/L,相对标准偏差为0.45%,线性范围为0~100μg/L。应用该方法检测了不同富硒酵母中的总硒含量为1.40~3.20mg/g其中,有机硒含量为1.30~2.97mg/g,加标回收率为95.4%~104.3%。 A method for the determination of organic and inorganic selenium in selenium-enriched yeast by hydride generation atomic fluorescence spectrometry (HG-AFS) has been developed. The effects of the instrumental and chemical parameters on atomic fluorescence signal of selenium were investigated, and the optimum conditions were recommended. The effects of digest reagents on digestion of samples and the interference from foreign ions on selenium were briefly discussed. The detection limit for selenium is 0. 0785 μg/L under the optimum condition. The linear range is 0~ 100 μg/L. The relative standard derivation is less than 0.45%. A total selenium content of 1.40~3.20 mg/g and the organic selenium content of 1.30~2.97 mg/g were detected respectively from the selenium enriched yeast by this method,and the recoveries were 95.4%~104.3%.
