摘要
目的:观察使用灵芝孢子粉后,癫痫点燃大鼠模型皮质和海马区脑源性神经营养因子(BDNF)的含量及神经元形态学的改变,探索癫痫发病机制以及灵芝孢子粉对癫痫的干预作用。方法:采用戊四氮(PTZ)腹腔注射制作W istar大鼠慢性点燃模型,并将其分为空白对照组、癫痫模型组和灵芝孢子粉组,点燃后断头取脑,免疫组化染色观察BDNF含量的变化及神经元形态学的改变。结果:所有大鼠均达到癫痫模型点燃标准。使用灵芝孢子粉的中药干预组与癫痫模型组相比,脑内BDNF含量明显升高,同时神经元形态学改变明显好转。结论:灵芝孢子粉能够增强脑源性神经营养因子的表达,从而减轻癫痫发作给神经系统带来的损伤,所以灵芝孢子粉可能具有减轻痫性发作、保护神经元的作用。
Objective: To observe the quantum variation and the neuronal morphologic changes of BDNF in epilepsy rat brain tissue after using Garoderma lucidum spores to study the mechanism of epilepsy and the influence of Garoderma lucidum spores upon the disease. Method: We used PTZ intraperitoneal injection to prepare epilepsy igniting models and divided them into 3 groups at random: 1. blank contrast group, 2. kindled epilepsy model group and 3. Garoderma lucidum spores group. The head was cut off after igniting and the brain tissue was taken out, and the neuronal changes were examined by immunohistochemistry stain. Results:All rats reached kindled standard. The BDNF content of Garoderma lucidum spores group was higher than that of kindled epilepsy model group, and at the same time, the neuronal morphology turned better. Conclusion: Garoderma lucidum spores can decreased the injury caused by epilepsy by strengthening the BDNF expression, so it seems to lessen epilepsy outbreak and protect neurons.
