摘要
目的通过比较CIK细胞和LAK细胞的增殖、表型和抗瘤效应,为临床应用CIK细胞过继免疫治疗提供资料。方法取12例恶性肿瘤患者外周血,常规分离成单个核细胞,用不同细胞因子培养诱导出LAK细胞和CIK细胞,观察两种细胞的增殖、表型和抗瘤效应。结果培养后12天,CIK细胞数量达原来的11.5倍,而LAK细胞数量只有原来的5.2倍;比较培养前后CIK细胞的表型,可发现CD3+、CD3+CD4+、CD3+CD8+、CD3+CD56+、CD25+在培养后均有明显增高(P<0.05或0.001)。CIK细胞杀伤K562细胞和Raji细胞的能力均明显优于LAK细胞(P<0.05或0.001)。结论CIK细胞不仅具有强大的增殖功能,并且对不同的肿瘤细胞系显示出较LAK细胞更强的杀伤活性。因此,它是一种新型的过继免疫治疗方法。
Objective To collect data for adoptive immunotherapy of malignant tumors by comparing of the proliferation, phenotype and the anti- tumor reaction between cytokine-induced killer (CIK) cells and LAK cells. Methods The peripheral blood samples were collected from 12 patients with malignant tumors. Then the cells were separated to single nuclear cells. Different cytokines were used to induce LAK cells and CIK cells. The observation of the cell proliferation, phenotype and anti - tumor reaction had been done. Results 12 days after the cell cultivation, the CIK cells had produced 11.5 times more than of the original, while the LAK cells had only produced 5.2 times more than of the original. BY comparing the CIK phenotype before and after cultivation, an increase of CD3^+, CD3^+CD4^+, CD3^+ CD8^+, CD3^+CD56^+and CD25^+ cells was found after cultivation ( P 〈0.05 or 0. 001). The ability of CIK cells in killing K562 cells and Raji cells was stronger than LAK cells ( P 〈 0.05 or 0. 001). Conclusion Not only can CIK cells have strong proliferate ability, but also have stronger killing ability in killing different tumor cell lines than LAK cells. It can be used as a'new type of adoptive immunotherapy.
