复方郁金胶囊抗抑郁抗焦虑的作用 被引量:24

摘要 目的:以抑郁和焦虑模型小鼠为实验对象,观察复方郁金胶囊的抗抑郁和抗焦虑作用。方法:实验于2004-10/2004-12在北京大学基础医学院药理学系神经精神药理实验室完成。选择雄性ICR小鼠250只,随机分成5组,每组50只,即对照组、复方郁金胶囊0.9,1.8,3.6g/kg3个剂量组、阳性药物组(丙咪嗪或地西泮),每组再均分为5组,分别进行小鼠强迫游泳、小鼠悬尾实验、小鼠自主活动、高台十字迷宫、孤养应激实验。复方郁金胶囊是将中药郁金、酸枣仁、合欢和茯苓的干燥水提取物,按30.5∶29.5∶32∶8的配比制成(样品由石家庄七月七生物高科技制品有限公司提供,批号:20041017)。评估方法和条件:①在强迫游泳实验中,观察小鼠在水中呈漂浮状态或仅有细小肢体运动以保持头部浮在水面的不动时间。②悬尾实验“不动时间”以计算机上小鼠活动曲线在6min内呈直线部分的累计之和。③在高台十字迷宫实验记录小鼠进入开臂的次数和停留时间。④在孤养小鼠应激实验采用电子体温计测定小鼠肛温变化,测定时体温计插入小鼠体内的深度和保留时间应一致。统计5组实验数据,分析复方郁金胶囊对小鼠的抗抑郁和抗焦虑作用。结果:实验小鼠250只全部进入结果分析。①强迫游泳实验结果:复方郁金胶囊1.8,3.6g/kg剂量组小鼠不动时间分别为70,48s,与对照组(130s)比较,差异有显著性意义(P<0.05)。②悬尾实验结果:复方郁金胶囊1.8,3.6g/kg剂量组小鼠不动时间比对照组明显缩短(49,48,86s,P<0.05)。③高台十字迷宫实验结果:复方郁金胶囊1.8,3.6g/kg剂量组小鼠在开臂内停留的时间和进入开臂内次数比对照组显著增加(74,66,37s;8.9,8.6,4.9次;P<0.05)。④孤养小鼠应激实验结果:复方郁金胶囊1.8,3.6g/kg剂量组小鼠体温分别升高0.42,0.37℃,对照组体温平均升高0.75℃;与对照组比较,差异有显著性意义(P<0.05)。⑤各组自主活动次数比较:复方郁金胶囊各剂量组小鼠不比对照组增加。结论:复方郁金胶囊在不增加小鼠自主活动基础上,可改善抑郁和焦虑模型小鼠行为,具有抗抑郁和抗焦虑作用。 AIM: To observe the antidepressant and anxiolytic effect of compound yujin capsule in mice models with depression and anxiety. METHODS: The experiment was conducted in Neural Pharmacology Laboratory of Department of Pharmacology, School of Basic Medical Sciences of Peking University from October to December 2004. A total of 250 male ICR mice were randomly divided into 5 groups: control group, 0.9, 1.8 and 3.6 g/kg yujin groups and positive drugs group (imipramine or diazepam) with 50 mice in each group, and then the 5 groups were subdivided into another 5 groups to perform the forced swimming; tail suspension, locomotor activity, elevated pulse-maze test and the stress induced hyperthermia in single-hoUsed mice, respectively. The yujin capsule was prepared with the dry extract of yujin, suanzaoren, hehuan and yujin with a proportion of 30.5:29.5:32:8 (the sample was provided by the Shijiazhuang Qiyueqi Biological High-Teeh Production Co., Ltd. No. 20041017). The evaluation methods and requirements: ①The immobility time, which referred to the time of lying on the surface of water or only having a mild movement of limbs with the head above the water of the mice, was measured in the forced swimming test. ②Immobility time in tail suspension test referred to the sum of horizontal part of the activity curve in 6 minutes on computer. ③In the elevated pulse-maze test, the number of open arm entering and keeping time were recorded. ④ The change of rectal temperature was measured with electronic clinical thermometer in the stress induced hyperthermia in single-housed mice test. The experimental statistics of the 5 groups were summed up to analyze the antidepressant and anxiolytic effect of compound yujin capsule on mice. RESULTS: All 250 mice were involved in the result analysis. ①In forced swimming experiment: The immobility time in 1.8 and 3.6 g/kg yujin groups was 70 and 48 seconds, respectively, which had significant differences compared with the control group (130 seconds, P 〈 0.05).② In tail suspension test: The immobility time in 1.8 and 3.6 g/kg yujin groups was shorter than the control group (49, 48 and 86 seconds, respectively, P 〈 0.05). ③In elevated pulse-maze test, the keeping time and number of open arm entering in 1.8 and 3.6 g/kg yujin groups were obviously increased compared with the control group (74, 66 and 37 seconds; 8.9, 8.6 and 4.9 times; P 〈 0.05). ④ In stress induced hyperthermia in singly-housed mice test: The average temperatures were increased 0.42 ℃ and 0.37℃, respectively in the 1.8, 3.6 g/kg yujin groups and the control group increased 0.75℃; there were significant differences among them (P 〈 0.05). ⑤The comparison of the locomotor activity times: It was not'increased in the yujin groups at different doses compared with the control group. CONCLUSION: The findings suggest that compound yujin capsule appears to be useful in the improvement of depression and anxiety in mice, without increase of locomotor activity.
