豌豆光系统I多肽HPCE分离特性的研究 被引量:5

ISOLATION OF PHOTOSYSTEM Ⅰ PROTEIN COMPLEX AND HPCE ANALYSIS OF THEIR POLYPEPTIDES

摘要 为进一步探讨豌豆光系统I(PSⅠ)的结构与多肽组成,本研究提取了经DOC-PAGE分离的光系统I多肽,采用毛细管凝胶电泳方法,以1%的葡聚糖为毛细管凝胶线性高分子筛分基质,0.5%乙基纤维素,0.1%SDS为磷酸盐缓冲液添加剂。在35min内进行了较好地分离。所得的色谱图基线平稳、分离度较高。为进一步利用毛细管电泳技术进行蛋白复合物多肽的分离奠定了良好的基础。 In order to discuss the structure and polypeptide composing of photosystem, polypeptides were extracted from photosystem Ⅰ DOC - PAGE separated from photosynthetic membranes in the leaves of Pisum sativum L.. A CGE method with 1% dextran as the sieving matrix and 0. 5% ethyl - cellulose, 0.1% SDS as the additive of the phosphate buffer was established for the separation of polypeptides obtained from photosynthetic membranes, baseline separation of the sample components could be realized in 35 rain with this method. A technical basement was established for the polypeptides separation of membrane protein with HPCE.
