摘要
为提高尿素包合法富集花生四烯酸的效率,通过正交设计对甲醇用量、结晶温度、尿/酯比、降温速度等因素进行分析,发现降温速度对分离效果有显著影响、结晶过程是制约分离效果的瓶颈。测定了油酸、亚油酸的尿素包合物的溶解度和超溶解度。当结晶温度为-7℃、降温速度5℃.h-1、脂肪酸甲酯∶尿素∶甲醇为1 g∶2 g∶12 mL时,花生四烯酸的质量分数为87.02%、收率为88.66%。
In the process of purification of arachidonic acid(AA) from fungi lipids by urea inclusion,orthogonal design were used. Crystallization temperature and speed of cooling-down had an outstanding effect on AA yield. So a crystallography study was carried out. Solubility and super-solubility curve were presented. When the temperature of crystallization was 7℃,the speed of cooling down was 5℃·h^-1 ,ratio of urea/fatty acid/ methanol was 1 g : 2 g : 13 mL, AA was separated with a purity of 87.02% (mass fraction)and recovery of 88.66%.
