HPLC法测定新疆贯叶连翘中绿原酸和芦丁的含量

Determination of Chlorogenic Acid and Rutin of Hypericum perforatum in Xinjiang by HPLC

采用液相色谱法测定贯叶连翘中绿原酸和芦丁含量。用Hyperil ODS色谱柱（12．5cm×6．0mmi．d．，5μm）进行分离。流动相为甲醇-1％冰醋酸梯度洗脱，流速为0．6mL／min。检测波长为343nm。绿原酸线性范围为0．0076～0．3040μg，相关系数R=0．9997。芦丁线性范围为0．312～3．120μg，相关系数R=0．9999。样品的平均回收率分别为101．2％，99．8％。此方法准确、快速，适用于绿原酸和芦丁的定量分析。

To develop a HPLC method for the determination of chlomgenic acid and turin in Hypericum perfomtum L in Xinjiang.Tne ehlorogenie acid and rutin were separated on a Hyperil ODS（12.5 cm× 6.0 mm i.d.,5μ m） with a mixture of methanol and 1% acetic acid as mobile phase, at a flow rate of 0.6 mL/min. Tne detection wavelength was 343 nm. Tne linearity of ehlorogenie acid was found in the image of 0.0076 ～ 0. 3040μg, R=0. 9997, the average recovery was 101.2 %, RSD = 0.88%. Tne linearity of rutin was 0. 312 ～3.120 μg, R = 0. 9999, the average recovery was 99.8%, RSD = 1.58%. This method is reliable, accurate and suitable for the determination of ehlorogenie acid and rutin.
