摘要
采用液相色谱法测定贯叶连翘中绿原酸和芦丁含量。用Hyperil ODS色谱柱(12.5cm×6.0mmi.d.,5μm)进行分离。流动相为甲醇-1%冰醋酸梯度洗脱,流速为0.6mL/min。检测波长为343nm。绿原酸线性范围为0.0076~0.3040μg,相关系数R=0.9997。芦丁线性范围为0.312~3.120μg,相关系数R=0.9999。样品的平均回收率分别为101.2%,99.8%。此方法准确、快速,适用于绿原酸和芦丁的定量分析。
To develop a HPLC method for the determination of chlomgenic acid and turin in Hypericum perfomtum L in Xinjiang.Tne ehlorogenie acid and rutin were separated on a Hyperil ODS(12.5 cm× 6.0 mm i.d.,5μ m) with a mixture of methanol and 1% acetic acid as mobile phase, at a flow rate of 0.6 mL/min. Tne detection wavelength was 343 nm. Tne linearity of ehlorogenie acid was found in the image of 0.0076 ~ 0. 3040μg, R=0. 9997, the average recovery was 101.2 %, RSD = 0.88%. Tne linearity of rutin was 0. 312 ~3.120 μg, R = 0. 9999, the average recovery was 99.8%, RSD = 1.58%. This method is reliable, accurate and suitable for the determination of ehlorogenie acid and rutin.
