呼吸链抑制剂对HepG2细胞DMT1表达量的影响

摘要 目的:研究缺氧与二价金属离子转运蛋白(d ivalent m etal transporter 1,DMT1)基因表达的关系,观察线粒体有氧呼吸链抑制剂对HepG2细胞DMT1表达量的影响。方法:将呼吸链抑制剂叠氮钠(NaN3)和鱼藤酮(rotenone)分别与HepG2细胞共孵育,用W estern B lot法检测DMT1的表达。结果:10 mmol/L和20 mmol/L的NaN3浓度可明显增加HepG2细胞±IRE DMT1的表达;20μmol/L的Rotenone可明显增加+IRE DMT1的表达,而对-IRE DMT1无明显影响。结论:DMT1的表达受呼吸链的调控,而+IRE DMT1与-IRE DMT1的表达调控可能不完全一样。 Objective: To investigate the relationship between hypoxia and the expression of divalent metal transporter 1 ( DMT1 ), we observed the effect of inhibitors of respiratory chain on DMT1 expression.Methods: HepG2 cells were exposed to sodium azide ( NaN3 ) or rotenone and the expression of DMT1 were detected by Western Blot. Results: When HepG2 cells were exposed to 10 mmol/L and 20 mmol/L NaN3 for 1 h, the expression of the two isoforms of DMT1 increased ; when HepG2 cells were exposed to 20 μmol/L rotenone, the expression of + IRE DMT1 increased, while the expression of - IRE DMT1 only changed littie, Conclusion: The expression of DMT1 may be regulated by the respiratory chain, and the expression of the two isoforms of DMT1 may be regulated differently.
