核转录因子ARNT对KLF4调控作用的初步研究 被引量:1

摘要 目的 探讨核转录因子ARNT和KLF4间可能存在的信号调控机制。方法构建ARNT真核表达质粒,并与其二聚体配体基因AHR表达质粒分别联合含KLF4启动子的荧光素酶报告载体体外共转染细胞,通过对荧光素酶活性检测分析ARNT对KLF4基因的调控作用。结果同空白对照比较,细胞中转染含ARNT质粒后,引起KLF4启动子荧光素酶活性增高2倍多。另一方面,共转染ARNT及其二聚体配体基因AHR,KLF4启动子荧光素酶活性较空白对照降低近1/8。结论ARNT可活化KLF4启动子,激活KLF4表达,而AHR共转染明显抑制KLF4启动子活性。 Objective The aim of study is to explore the signal regulation pathway between Krüppel - like Factor 4 (KLF4) and aryl hydrocarbon nuclear translocator factor (ARNT). Methods ARNT plasmid was constructed and cotransfected in vitro with KLF4-1uciferase reporter vector as well as AHR vector. Results The results of luciferase assay revealed that ARNT vector transfection could increase value of KLF4 to 2 folds, while cotransfected AHR vector could decrease luciferase activities to 8 times greatly. Conclusion ARNT could enhance activity of Klf4 promoter, while AHR compete to combine and antagonize Klf4 promoter activation.
